PCR Packaging Procedures

In polymerase chain reaction (PCR) experiments, the packaging of reagents and samples directly impacts the accuracy and safety of experimental results. Standard PCR packaging procedures must strictly adhere to the principles of sterility, contamination prevention, and clear labeling. Specific procedures are outlined below.


1. Preparation

Before proceeding, wear a lab coat, gloves, and a mask to ensure a clean environment (a clean bench or biosafety cabinet is recommended). Check that all consumables (e.g., centrifuge tubes, PCR plates) are free of DNase/RNase contamination and use autoclaved or enzyme-free sterilizers. Reagents (e.g., primers, template DNA, enzymes, etc.) should be aliquoted into individual vials to avoid repeated freeze-thaw cycles.

 

2. Aliquoting and Labeling

Aliquoting Reagents: Use a pipette to dispense master mix, primers, etc. into centrifuge tubes or PCR plate wells according to experimental needs, accurately dispensing each volume to the nearest microliter. Use disposable pipette tips to avoid cross-contamination.

Labeling Standards: All containers must be clearly labeled with the name, concentration, date, and operator number. Waterproof labels or direct engraving are recommended to prevent illegible markings.


3. Packaging and Sealing

After aliquoting, immediately tighten the centrifuge tube caps or seal the PCR plates (using heat-sealing film or specialized adhesive film) to prevent leakage. For long-term storage, it is recommended to place the packaging in a sealed bag and add a desiccant (such as silica gel beads) to maintain a dry environment.


4. Storage and Transportation

After packaging, PCR reagents should be stored separately at -20°C (long-term) or 4°C (short-term) to avoid temperature fluctuations. Transport should be performed in an insulated container filled with cushioning material to prevent shock and leakage. For biohazardous samples, compliance with biosafety packaging requirements under UN 3373 or relevant regulations is required.

Standardized PCR packaging procedures are essential for experimental reliability. Strict aseptic control and clear labeling can effectively reduce contamination risks and ensure data accuracy.
